Mazdutide 10 mg vial dosage
A 10 mg Mazdutide vial mixed with 1 mL of bacteriostatic water gives 10 mg/mL, so 6 mg is 60 units (0.6 mL) on a U-100 insulin syringe. At 6 mg a dose, one vial holds 1 dose.

What Mazdutide is
- Class:
- Dual GLP-1/glucagon receptor agonist, a peptide, co-developed by Innovent Biologics and Eli Lilly
- Also called:
- IBI362, LY3305677
- Used for:
- Approved in China for chronic weight management and for glycemic control in type 2 diabetes. Not FDA-approved in the United States, where it is sold to consumers as a research-chemical vial.
- Human evidence:
- Moderate human evidence. A US-based, multicentre, placebo-controlled phase 2 trial in adults with obesity or overweight reported dose-ranging efficacy and safety data supporting later-stage development. Mazdutide has since been approved by China's National Medical Products Administration (NMPA) for chronic weight management and for glycemic control in type 2 diabetes, each following its own phase 3 programme in Chinese adults - real regulatory review, but not an FDA review, and I could not verify the phase 3 trial results directly against a peer-reviewed publication for every claim in Innovent's own announcements.
- US status:
- Research use only. Not FDA-approved for any indication in the United States and not on the FDA's approved-drug list; sold to US consumers as a research chemical. China's NMPA approved mazdutide for chronic weight management in June 2025 and for glycemic control in type 2 diabetes in September 2025, per Innovent Biologics' own regulatory announcements. I did not verify approval status in any other jurisdiction, including Australia.

Supplies for one cycle with 10 mg vials
Counted for the low and high ends of the common protocol, mixing each vial with 1 mL of water. One new syringe per injection.
| Plan | Injections | 10 mg vials | Insulin syringes | Bacteriostatic water | Alcohol swabs |
|---|---|---|---|---|---|
| 3 mg, 1×/week, 24 weeks | 24 | 8 | 24 | 8 mL (1 × 10 mL bottle) | 64 (1 × 100-pack) |
| 9 mg, 1×/week, 24 weeks | 24 | 22 | 24 | 22 mL (3 × 10 mL bottles) | 92 (1 × 100-pack) |
Arithmetic on the protocol and this vial size. Swabs: two per vial mixed (both stoppers) and two per injection (vial stopper and skin). An opened multi-dose vial has a use-by date (see storage), which on a slow schedule can matter more than the dose count. Plan your own numbers with the protocol builder.
How to mix a 10 mg Mazdutide vial
For this vial: 1 mL of bacteriostatic water makes 10 mg/mL, so 6 mg is 60 units (0.6 mL) on a U-100 insulin syringe. Pick a different volume from the units table if you want bigger or smaller draws; the number of doses stays the same.
- Wipe the rubber stoppers of both the peptide vial and the bacteriostatic water vial with an alcohol swab and let the alcohol dry. [3][4]
- With a new sterile needle and syringe, draw up the amount of bacteriostatic water you have chosen for the dilution. [3][4][5]
- Inject the water into the peptide vial by aiming the stream against the inside glass wall, which helps prevent foaming. [6]
- Swirl or roll the vial gently until the powder has fully dissolved; do not shake it. Do not inject the solution if it is cloudy or has particles in it. [3][4][6]
- Label the vial with the date and time it was mixed, the amount of water added and how long it can be used after mixing. [3]
- Store the mixed vial in the refrigerator at 2-8°C (36-46°F) and do not freeze it. [6]
Step by step with pictures: how to reconstitute peptides. About the water itself: bacteriostatic water.
Storing Mazdutide vials
- Freeze-dried (lyophilized) peptide keeps far longer than peptide in solution; peptide suppliers advise storing the powder at -20°C away from bright light. [7]
- Protect peptide vials from heat and light. [7][8]
- Avoid repeated freeze-thaw cycles; a prescription peptide label also says not to freeze a vial once it has been mixed. [6][7]
- Once a multi-dose vial, such as bacteriostatic water, has been opened (needle-punctured), date it and discard it within 28 days unless its manufacturer states another date. [9][10]
More detail: peptide storage and shelf life.
Injecting safely
- Clean the injection site with an alcohol swab and let the skin dry completely before injecting. [11][12]
- Pinch up a fold of skin and fat (not muscle) and insert the needle at a 45- to 90-degree angle: 90 degrees where there is enough fatty tissue, 45 degrees where there is little. [11][12][13]
- Pulling back on the plunger (aspiration) is not necessary for a subcutaneous injection, because the chance of the needle being in a blood vessel is small. [11]
- Inject slowly, at roughly 10 seconds per mL. [11]
- Common sites are the belly, the thigh and the upper arm; rotate sites, keeping each injection at least 1 inch (2.5 cm) from the last. [6][11][12]
Hygiene and warning signs
- Use a new sterile needle and syringe for every injection and every time you enter a vial; never reuse them. [5][9][12]
- Never share needles, syringes or pens with anyone, even if the needle is changed; unsafe injection practices spread hepatitis B, hepatitis C and HIV. [5][8][9]
- Put used needles and syringes in a sharps disposal container straight away (FDA-cleared if possible), never loose in the trash, recycling or toilet, and dispose of the container following your community's guidelines. [14][15]
- Contact a health care provider if the injection site shows signs of infection, such as redness, swelling, red streaks or a fever. [12]
Where to inject: subcutaneous injection sites.
How Mazdutide works
- Mazdutide (trial codes IBI362 and LY3305677) is a peptide given as a once-weekly injection that activates two receptors at once: the GLP-1 receptor and the glucagon receptor. It was developed for obesity and type 2 diabetes. [16]
- A 2026 patent review groups mazdutide with related oxyntomodulin analogues and traces its original peptide design to Eli Lilly, with later formulation and clinical development led by Innovent Biologics. Oxyntomodulin is a gut hormone that itself activates both the glucagon and GLP-1 receptors. [17][18]
- The GLP-1 receptor is the target of GLP-1-only drugs already used for type 2 diabetes and obesity. The case for adding glucagon-receptor activity rests on emerging evidence that glucagon helps regulate energy balance and fat metabolism as well as blood glucose; a 2026 review describes this extra benefit as possible rather than proven. [19]
What the Mazdutide research found
Human evidence: moderate human evidence. Each result says what was studied.
- Human studyIn a US phase 2 randomised trial of 179 adults with obesity or overweight and no diabetes, body weight fell by 7.3% in the 3-6 mg group (on 3 mg until week 32), 15.6% on 10 mg and 18.1% on 16 mg weekly at 32 weeks, against 0.9% on placebo. Further weight loss was seen by 48 weeks. [20]
- Human studyIn GLORY-1, a phase 3 randomised trial of 610 Chinese adults with obesity or overweight, body weight changed by -11.00% on 4 mg and -14.01% on 6 mg at 48 weeks, against +0.30% on placebo. At least 15% weight loss was reached by 35.7%, 49.5% and 2.0% respectively. [21]
- Human studyIn GLORY-2, a 60-week phase 3 trial in Chinese adults with a BMI of 30 or more (461 treated), 9 mg weekly lowered body weight by 16.65%, against 1.50% on placebo. [22]
- Human studyIn a phase 3 trial of 320 Chinese adults whose type 2 diabetes was not controlled by diet and exercise alone, HbA1c fell by 1.57% on 4 mg and 2.15% on 6 mg at 24 weeks, against 0.14% on placebo, with weight loss of 5.61% and 7.81% against 1.26%. [23]
- Human studyIn a 28-week phase 3 trial of 731 Chinese adults with type 2 diabetes on oral medicines, both 4 mg and 6 mg mazdutide lowered HbA1c more than 1.5 mg dulaglutide (differences of 0.24% and 0.30%) and cut body weight by a further 3.78% and 5.76%. [24]
Mazdutide side effects and risks
Mazdutide has no FDA label. The safety data come from phase 1-3 trials funded by Innovent Biologics or Eli Lilly, most of them in Chinese adults.
- Human studyGastrointestinal effects are the most common adverse events across the trials. In GLORY-2 (9 mg), vomiting affected 53.1%, nausea 46.9% and diarrhoea 39.4% of participants, against 1.3%, 3.2% and 6.5% on placebo; most were mild to moderate. [22]
- Human studyIn the US phase 2 trial, stopping treatment because of adverse events was most common on 16 mg (20%), mainly because of gastrointestinal effects. In GLORY-1 the rates were 1.5% on 4 mg and 0.5% on 6 mg, against 1.0% on placebo. [20][21]
- Human studyIn the type 2 diabetes trial against dulaglutide, diarrhoea, nausea and vomiting were the most common adverse events, and gastrointestinal events were more frequent on mazdutide than on dulaglutide. [24]
- Human studyIn an early 12-week dose-escalation study of 36 Chinese adults, three people on mazdutide had mild, symptom-free heart findings picked up on ECG. No one stopped for safety reasons and there were no serious adverse events. [16]
- Human studyA 2026 review of glucagon/GLP-1 dual agonists reports that mazdutide and survodutide generally raised heart rate by about as much as GLP-1-only drugs. Their cardiovascular effects are still being established drug by drug. [19]
Other things to know
- Not FDA-approved in the United States. It is approved in China, but a foreign approval does not transfer to the US supply chain, and a US-sold "research" vial carries no FDA review of its identity, purity or dose.
- As a dual GLP-1/glucagon agonist, it shares the class-wide gastrointestinal adverse-event profile (nausea, vomiting, diarrhoea) reported in its own phase 2 and phase 3 trials, generally more common at higher doses.
- The added glucagon-receptor activity is a different mechanism from a GLP-1-only drug, and long-term human safety data outside its own trial programme and China's post-approval surveillance are limited.
- Vials sold outside the NMPA-approved supply chain, including anything marketed to US buyers, are not the reviewed product and carry no verified content or concentration.

Questions
- How much water do I add to a 10 mg Mazdutide vial?
- There is no single correct amount: water changes how many units a dose is, not how many doses the vial holds. With 1 mL the vial is 10 mg/mL and 6 mg is 60 units (0.6 mL) on a U-100 insulin syringe. The units table gives 1, 1.5 mL.
- How many doses are in a 10 mg Mazdutide vial?
- 3 doses at 3 mg, 1 dose at 6 mg, 1 dose at 9 mg.
- How long does a 10 mg Mazdutide vial last?
- At 6 mg a dose and once weekly, about 1 weeks. How long a mixed vial keeps is a separate limit; see storage below.
- How many 10 mg vials do I need for a cycle?
- 8 vials at 3 mg 1 time a week for 24 weeks; 22 vials at 9 mg 1 time a week for 24 weeks.
- What does 1 unit equal in a 10 mg Mazdutide vial?
- 100 mcg with 1 mL, 66.67 mcg with 1.5 mL of bacteriostatic water, on a U-100 syringe.
