Mazdutide Dosage: Trial Doses, Common Protocol and Calculator
Mazdutide dosage: doses from its China phase 3 trials, and the wider range in circulation in the research-chemical market. No FDA approval, no FDA-established dose.
The short version
Mazdutide has no FDA-approved dose because it has no FDA-approved product. Its China phase 3 trials tested 4 mg and 6 mg weekly for weight management, and separately 9 mg weekly. The research-chemical market uses a wider 3-9 mg weekly range, with a step-up titration over about 16 weeks. That is common practice, not trial-established, and only two vendor-side sources report it, which do not fully agree.
- China's phase 3 trials used 4 mg or 6 mg weekly (GLORY-1) and 9 mg weekly (GLORY-2).
- Research-market sources describe a 3-9 mg titration, once weekly. It is not trial-established, and the two sources found do not fully agree.
- No FDA review has happened for any dose, in any indication.
- Use a calculator for reconstitution and injection-volume math, not a guess.
Mazdutide has no FDA-approved dose, because it has no FDA-approved product in the United States. This page reports trial doses from its published and announced China programme, and separately what circulates in the research-chemical market. Neither is a recommendation. For approval status and mechanism, see the main mazdutide guide.
US legal status - Research use only
No FDA-approved product exists for mazdutide, and no dose has been reviewed by the FDA.
Mazdutide dosage: the common protocol
The research-chemical market uses a step-up titration from around 3 mg to a 9 mg weekly maintenance dose, injected subcutaneously once weekly. The titration takes about 16 weeks inside a roughly 24-week cycle. This is common practice, not a dose any trial or regulator has established, and the two sources behind it do not fully agree.
Common protocol
What is commonly used, not a dose established in trials. No regulator has set a dose for Mazdutide. Sources differ, so the ranges below span them.
- Dose per administration
- 3-9 mg
- How often
- once weekly, titrated upward from a lower starting dose
- Route
- Subcutaneous injection
- Cycle length
- 24 weeks
- Break between cycles
- 8-12 weeks
- Common vial size
- 10 mg
- Timing
- Same day each week; not tied to meals.
Sources describe different targets within the same 3-9 mg weekly range used in mazdutide's own China trials: one titrates to a 9 mg maintenance dose over 16 weeks, the other reports a fixed 4-6 mg weekly maintenance range without a titration schedule. Neither source is a clinic or prescriber protocol - mazdutide is not FDA-approved and is not sold through a US telehealth pathway.
Plan a cycle in the protocol builder or work out injection volume in the dosage calculator.
The common Mazdutide dose is 3-9 mg, once weekly, titrated upward from a lower starting dose. From a 10 mg vial mixed with 1 mL of bacteriostatic water, 6 mg is 60 units (0.6 mL) on a U-100 insulin syringe.

Units are on a standard U-100 insulin syringe (100 units = 1 mL), rounded to the nearest half unit - half-unit marks are on 0.3 mL syringes. Water volumes are examples: more water means more units per dose, not more doses per vial. For any other vial or dose, use the calculator below.

Doses used in mazdutide’s own trials
A US-based, multicentre, placebo-controlled phase 2 trial Hsia 2026 - Hsia SH, Bays HE, Billings LK et al. Efficacy and safety of mazdutide in adults with obesity or overweight: a US-based, multicentre, phase 2, randomised, placebo-controlled clinical trial. Lancet Diabetes Endocrinol. 2026., in the references below tested a dose-ranging series in adults with obesity or overweight. China’s phase 3 programme has since reported:
| Trial | Population | Dose(s) | Duration |
|---|---|---|---|
| GLORY-1 (phase 3) | Chinese adults, obesity/overweight | 4 mg or 6 mg once weekly | 48 weeks |
| GLORY-2 (phase 3) | Chinese adults, moderate-to-severe obesity | 9 mg once weekly | 60 weeks |
| Type 2 diabetes phase 3 | Chinese adults with T2D | 4 mg or 6 mg once weekly | Reported against dulaglutide Guo 2026 - Guo L, Zhang B, Xue X et al. Mazdutide versus dulaglutide in Chinese adults with type 2 diabetes. Nature. 2026;652(8108):181-188., in the references below |
A trial protocol assigned these doses to monitored participants under clinical supervision. They are not a recommendation for anyone taking an unlabelled vial outside that system.
How to reconstitute mazdutide
The injection volume for any target amount depends on how a specific vial was reconstituted (diluent volume and resulting concentration), which this page cannot know. The dosage calculator works through that arithmetic. It does not choose a dose.
What is not established
- No FDA-approved dose exists, because no US product has completed FDA review.
- The research-market titration above is common practice, reported by two sources that do not fully agree. No trial or regulator has established it.
- China’s exact approved dosing on its own product label has not been verified against a Chinese-language source. The figures here come from trial data and Innovent’s own English-language announcements.
Who should not use it
No FDA-reviewed contraindication list exists for mazdutide in the US, because no such review has happened. Its trials excluded people with significant medical conditions under close monitoring. That shows who a study considered eligible, not that anyone else is safe to use it. Nobody should self-direct an unapproved research chemical around a current medical condition, medication or pregnancy.
Mazdutide side effects and risks
Its phase 2 and phase 3 trials report gastrointestinal effects (nausea, vomiting, diarrhoea) as the most common adverse events, generally more frequent at higher doses, as with other GLP-1-class drugs. That is trial data from monitored populations and says nothing about long-term use outside that system. A vial bought outside a reviewed supply chain carries no verified content, purity or actual concentration.

Common questions
What is the usual mazdutide dose?
Research-market sources most often describe a step-up titration from around 3 mg to a 9 mg weekly maintenance dose, injected subcutaneously once weekly. This is common practice reported by peptide vendor and blog pages, not a dose established by any trial or regulator. The two sources found do not fully agree. See "The common protocol" below.
What dose did mazdutide's own trials use?
A US phase 2 trial and China-based phase 3 trials tested several doses. GLORY-1, the first completed phase 3 weight-management trial, tested 4 mg and 6 mg once weekly. GLORY-2 tested 9 mg once weekly in people with moderate-to-severe obesity. A separate phase 3 diabetes trial tested 4 mg and 6 mg once weekly.
Is there an FDA-approved mazdutide dose?
No. Mazdutide has no FDA-approved product. China's NMPA has approved it for two indications, each with its own approved dosing, which I have not verified against a Chinese-language label. The figures here come from published trial data and research-market sources, not FDA review.
Why do the two dosing sources disagree?
One source describes a step-up titration to a 9 mg weekly maintenance dose over about 16 weeks. The other describes a fixed 4-6 mg weekly range with no titration, closer to the GLORY-1 trial doses. Neither is a clinic or prescriber protocol. The disagreement is a reason for caution, not a reason to average the two into one number.
How do I work out reconstitution and injection volume?
That is arithmetic specific to a vial's concentration and diluent volume, which this page cannot know for you. Use the dosage calculator to work through the math for your vial and target amount. It performs the calculation. It does not choose a dose.
What are the main risks?
Its trials report gastrointestinal effects (nausea, vomiting, diarrhoea) as the most common adverse events, more frequent at higher doses. Beyond its trial populations and China's post-approval surveillance, there is no FDA-reviewed safety label. A vial bought outside a reviewed supply chain has no verified content or purity.
