CJC-1295 5 mg vial dosage

A 5 mg CJC-1295 vial mixed with 1.5 mL of bacteriostatic water gives 3.33 mg/mL, so 200 mcg is 6 units (0.06 mL) on a U-100 insulin syringe. At 200 mcg a dose, one vial holds 25 doses.

What CJC-1295 is

Class:
Synthetic GHRH (1-29) analogue
Also called:
CJC-1295 with DAC, CJC-1295 without DAC, Modified GRF (1-29), mod GRF 1-29, Tetrasubstituted GRF(1-29)
Used for:
Investigated as a long-acting growth hormone-releasing hormone analogue to raise GH and IGF-I. Reached Phase 2 and was discontinued; never approved for any indication.
Human evidence:
Limited human data. Human pharmacokinetic data exists for the DAC form only, from two randomised, placebo-controlled, double-blind ascending-dose trials in healthy adults aged 21 to 61, reported in one 2006 paper. Those are surrogate hormone measures, not clinical outcomes, and development was discontinued at Phase 2. The without-DAC form (mod GRF 1-29) has no published human pharmacokinetic study I could locate. One entry, two very different evidence bases.
Half-life:
DAC form: an estimated 5.8–8.1 days in healthy adults, from two randomised, placebo-controlled trials. Without-DAC form (mod GRF 1-29): not established in humans; no published human PK study located.
US status:
Research use only. Not FDA-approved for any indication in the US. Reached Phase 2 clinical trials and was discontinued. Sold as a research chemical and not legally marketed for human use. Named explicitly in the WADA 2026 Prohibited List under S2.2.4 (growth hormone releasing factors), prohibited at all times.

Arithmetic on the protocol and this vial size. Swabs: two per vial mixed (both stoppers) and two per injection (vial stopper and skin). An opened multi-dose vial has a use-by date (see storage), which on a slow schedule can matter more than the dose count. Plan your own numbers with the protocol builder.

How to mix a 5 mg CJC-1295 vial

For this vial: 1.5 mL of bacteriostatic water makes 3.33 mg/mL, so 200 mcg is 6 units (0.06 mL) on a U-100 insulin syringe. Pick a different volume from the units table if you want bigger or smaller draws; the number of doses stays the same.

  1. Wipe the rubber stoppers of both the peptide vial and the bacteriostatic water vial with an alcohol swab and let the alcohol dry. [4][5]
  2. With a new sterile needle and syringe, draw up the amount of bacteriostatic water you have chosen for the dilution. [4][5][6]
  3. Inject the water into the peptide vial by aiming the stream against the inside glass wall, which helps prevent foaming. [7]
  4. Swirl or roll the vial gently until the powder has fully dissolved; do not shake it. Do not inject the solution if it is cloudy or has particles in it. [4][5][7]
  5. Label the vial with the date and time it was mixed, the amount of water added and how long it can be used after mixing. [4]
  6. Store the mixed vial in the refrigerator at 2-8°C (36-46°F) and do not freeze it. [7]

Step by step with pictures: how to reconstitute peptides. About the water itself: bacteriostatic water.

Storing CJC-1295 vials

  • Freeze-dried (lyophilized) peptide keeps far longer than peptide in solution; peptide suppliers advise storing the powder at -20°C away from bright light. [8]
  • Protect peptide vials from heat and light. [8][9]
  • Avoid repeated freeze-thaw cycles; a prescription peptide label also says not to freeze a vial once it has been mixed. [7][8]
  • Once a multi-dose vial, such as bacteriostatic water, has been opened (needle-punctured), date it and discard it within 28 days unless its manufacturer states another date. [10][11]

More detail: peptide storage and shelf life.

Injecting safely

  • Clean the injection site with an alcohol swab and let the skin dry completely before injecting. [12][13]
  • Pinch up a fold of skin and fat (not muscle) and insert the needle at a 45- to 90-degree angle: 90 degrees where there is enough fatty tissue, 45 degrees where there is little. [12][13][14]
  • Pulling back on the plunger (aspiration) is not necessary for a subcutaneous injection, because the chance of the needle being in a blood vessel is small. [12]
  • Inject slowly, at roughly 10 seconds per mL. [12]
  • Common sites are the belly, the thigh and the upper arm; rotate sites, keeping each injection at least 1 inch (2.5 cm) from the last. [7][12][13]

Hygiene and warning signs

  • Use a new sterile needle and syringe for every injection and every time you enter a vial; never reuse them. [6][10][13]
  • Never share needles, syringes or pens with anyone, even if the needle is changed; unsafe injection practices spread hepatitis B, hepatitis C and HIV. [6][9][10]
  • Put used needles and syringes in a sharps disposal container straight away (FDA-cleared if possible), never loose in the trash, recycling or toilet, and dispose of the container following your community's guidelines. [15][16]
  • Contact a health care provider if the injection site shows signs of infection, such as redness, swelling, red streaks or a fever. [13]

Where to inject: subcutaneous injection sites.

How CJC-1295 works

  • CJC-1295 is a modified version of GRF(1-29), the 29-amino-acid active fragment of growth hormone-releasing hormone. It carries four amino acid substitutions plus an extra lysine at the C-terminus bearing a maleimidopropionamide group, which is what lets it bind albumin. [17]
  • The maleimide group binds covalently to a free thiol on circulating albumin (cysteine 34). In rats, CJC-1295 was found attached to albumin within 15 minutes of injection and was still present in plasma beyond 72 hours, and in the lab, albumin-bound versions were more stable against breakdown by the enzyme DPP-IV. [17]
  • Like natural GHRH, it activates the GHRH receptor on the anterior pituitary to release growth hormone. In rats, one injection gave four times the growth hormone exposure over two hours of unmodified GRF(1-29). [17]
  • In healthy adults, the estimated half-life of this albumin-binding CJC-1295 was 5.8 to 8.1 days. [18]

What the CJC-1295 research found

Human evidence: limited human data. Each result says what was studied.

  • Human studyIn two randomised, double-blind, placebo-controlled trials in healthy adults aged 21 to 61, a single injection raised average growth hormone 2- to 10-fold for 6 days or more and IGF-I 1.5- to 3-fold for 9 to 11 days, with larger rises at higher doses. After 2 or 3 doses given weekly or every two weeks, IGF-I stayed above baseline for up to 28 days. [18]
  • Human studyIn healthy men aged 20 to 40, one week after a single 60 or 90 mcg/kg injection, growth hormone was still released in pulses of unchanged frequency and size, but trough levels rose 7.5-fold, mean growth hormone by 46% and IGF-I by 45%. The two doses gave similar results. [19]
  • Animal studyIn mice unable to make their own GHRH, 5 weeks of daily CJC-1295 gave normal body weight and length. The same dose every 48 or 72 hours improved growth over placebo but did not fully normalise it. [20]
  • Human studyA 12-week Phase 2 randomised, placebo-controlled trial of CJC-1295 in people with HIV-associated visceral obesity (listed enrolment 120) was registered by ConjuChem in 2005 and is recorded as terminated in 2006. No results were posted. [21]

CJC-1295 side effects and risks

No trial has published systematic human safety data for CJC-1295 beyond short studies in healthy volunteers, and none for the form sold without DAC.

  • Human studyIn the two trials in healthy adults, which followed participants for 28 and 49 days, no serious adverse reactions were reported. The authors described CJC-1295 as relatively well tolerated, particularly at 30 or 60 mcg/kg. [18]
  • Regulator reviewFDA states that it has identified serious adverse events associated with CJC-1295, including increased heart rate and a systemic vasodilatory reaction, that available clinical data are limited, and that compounded versions may pose a risk of immunogenicity (an immune reaction to the drug). [22]

Other things to know

  • The with-DAC and without-DAC forms are sold under one name but differ structurally, and the albumin-binding linker is the reason the DAC form persists far longer. Buying "CJC-1295" without knowing which form is in the vial means not knowing whether the signal is short-lived or lasts days.
  • Development was halted at Phase 2 and the compound was never approved. There is no long-term human safety data for either form.
  • Sold as a research chemical, so identity, purity and which form you actually have are unverified.
  • Prohibited at all times in sport (WADA S2.2.4).
