Kisspeptin 10 mg vial dosage
A 10 mg Kisspeptin vial mixed with 5 mL of bacteriostatic water gives 2 mg/mL, so 150 mcg is 7.5 units (0.08 mL) on a U-100 insulin syringe. At 150 mcg a dose, one vial holds 66 doses.

What Kisspeptin is
- Class:
- Neuropeptide - KISS1 gene product, upstream regulator of GnRH secretion
- Also called:
- Kisspeptin-10, Kisspeptin-54, KP-10, KP-54, Metastin
- Used for:
- Studied in hospital-based human research as a reproductive-hormone signal - it stimulates LH and FSH release upstream of GnRH - and, in a small number of academic trials, for its effect on sexual and romantic brain processing. Sold to consumers for sexual desire and fertility use, a use no regulator has evaluated as safe or effective.
- Human evidence:
- Limited human data. FDA's 2024 evaluation of kisspeptin-10 for compounding found insufficient evidence to conclude it is effective for secondary hypogonadism in men, citing small, exploratory, short-duration studies with inconsistent dosing routes. The sexual-brain-processing findings come from a handful of single-dose, placebo-controlled academic trials in healthy volunteers, not patients, and have not been replicated as a treatment outcome. Human data exist and are genuine; they are small, mostly acute, and none support a treatment claim yet.
- Half-life:
- About 3.8 (±0.3) minutes for kisspeptin-10 by IV bolus in healthy men - the only human figure FDA identifies in its review of the published pharmacokinetic studies. For kisspeptin-54, a 2005 study measured a plasma half-life of about 27.6 minutes in six healthy men during a 90-minute IV infusion.
- US status:
- Research use only. No USP/NF monograph exists for kisspeptin-10 and it is not a component of any FDA-approved drug. In its October 2024 briefing document for the Pharmacy Compounding Advisory Committee, FDA proposed NOT adding kisspeptin-10 to the 503A Bulks List, citing incomplete physicochemical characterisation, unresolved immunogenicity risk for the injectable route, and insufficient evidence of effectiveness for secondary hypogonadism in men. That is a staff proposal tied to one nominated use, not a ban on the molecule, and not a determination for any other use. Kisspeptin does not appear in Australia's current Poisons Standard, so I have not verified a scheduling status there.

Arithmetic on the protocol and this vial size. Swabs: two per vial mixed (both stoppers) and two per injection (vial stopper and skin). An opened multi-dose vial has a use-by date (see storage), which on a slow schedule can matter more than the dose count. Plan your own numbers with the protocol builder.
How to mix a 10 mg Kisspeptin vial
For this vial: 5 mL of bacteriostatic water makes 2 mg/mL, so 150 mcg is 7.5 units (0.08 mL) on a U-100 insulin syringe. Pick a different volume from the units table if you want bigger or smaller draws; the number of doses stays the same.
- Wipe the rubber stoppers of both the peptide vial and the bacteriostatic water vial with an alcohol swab and let the alcohol dry. [4][5]
- With a new sterile needle and syringe, draw up the amount of bacteriostatic water you have chosen for the dilution. [4][5][6]
- Inject the water into the peptide vial by aiming the stream against the inside glass wall, which helps prevent foaming. [7]
- Swirl or roll the vial gently until the powder has fully dissolved; do not shake it. Do not inject the solution if it is cloudy or has particles in it. [4][5][7]
- Label the vial with the date and time it was mixed, the amount of water added and how long it can be used after mixing. [4]
- Store the mixed vial in the refrigerator at 2-8°C (36-46°F) and do not freeze it. [7]
Step by step with pictures: how to reconstitute peptides. About the water itself: bacteriostatic water.
Storing Kisspeptin vials
- Freeze-dried (lyophilized) peptide keeps far longer than peptide in solution; peptide suppliers advise storing the powder at -20°C away from bright light. [8]
- Protect peptide vials from heat and light. [8][9]
- Avoid repeated freeze-thaw cycles; a prescription peptide label also says not to freeze a vial once it has been mixed. [7][8]
- Once a multi-dose vial, such as bacteriostatic water, has been opened (needle-punctured), date it and discard it within 28 days unless its manufacturer states another date. [10][11]
More detail: peptide storage and shelf life.
Injecting safely
- Clean the injection site with an alcohol swab and let the skin dry completely before injecting. [12][13]
- Pinch up a fold of skin and fat (not muscle) and insert the needle at a 45- to 90-degree angle: 90 degrees where there is enough fatty tissue, 45 degrees where there is little. [12][13][14]
- Pulling back on the plunger (aspiration) is not necessary for a subcutaneous injection, because the chance of the needle being in a blood vessel is small. [12]
- Inject slowly, at roughly 10 seconds per mL. [12]
- Common sites are the belly, the thigh and the upper arm; rotate sites, keeping each injection at least 1 inch (2.5 cm) from the last. [7][12][13]
Hygiene and warning signs
- Use a new sterile needle and syringe for every injection and every time you enter a vial; never reuse them. [6][10][13]
- Never share needles, syringes or pens with anyone, even if the needle is changed; unsafe injection practices spread hepatitis B, hepatitis C and HIV. [6][9][10]
- Put used needles and syringes in a sharps disposal container straight away (FDA-cleared if possible), never loose in the trash, recycling or toilet, and dispose of the container following your community's guidelines. [15][16]
- Contact a health care provider if the injection site shows signs of infection, such as redness, swelling, red streaks or a fever. [13]
Where to inject: subcutaneous injection sites.
How Kisspeptin works
- The KISS1 gene makes a 145-amino-acid protein that is cut into several kisspeptins, including kisspeptin-54, -14, -13 and -10. All of them share the same ten-amino-acid tail, which is kisspeptin-10, the shortest form with full activity. [17][18]
- Kisspeptin activates the GPR54 receptor (also called KISS1R), which is concentrated in the hypothalamus. Kisspeptin neurons project to the GnRH neurons, and activating the receptor increases the pituitary's release of LH and FSH, which in turn raises sex-hormone output from the gonads. [17]
- The receptor is essential for normal puberty. In 2003, loss-of-function mutations in GPR54 were found to cause hypogonadotropic hypogonadism in humans, in a family with five affected siblings and separately in other patients. [19][20]
- Kisspeptin-10 clears in minutes: its plasma half-life, measured during and after an intravenous infusion, is about 3.8 minutes in healthy men. Kisspeptin-54 lasts longer, with a half-life of 27.6 minutes measured during infusion in men. [17][21]
What the Kisspeptin research found
Human evidence: limited human data. Each result says what was studied.
- Human studyIn a 2005 double-blind, placebo-controlled crossover study of 6 healthy men, a 90-minute intravenous infusion of kisspeptin-54 raised LH, FSH and testosterone compared with saline. [21]
- Human studyIn healthy men given intravenous kisspeptin-10, LH rose with dose up to 1 microgram/kg, and 3 micrograms/kg gave a smaller response. A 22.5-hour infusion in 4 men raised testosterone from 16.6 to 24.0 nmol/L. [18]
- Human studyIn a 2009 randomised double-blind trial, women with hypothalamic amenorrhoea were given twice-daily subcutaneous kisspeptin-54 or saline (5 per group) for 2 weeks. The strong LH and FSH rise seen on day 1 had largely disappeared by day 14 (tachyphylaxis). [22]
- Human studyIn a 2015 phase 2 open-label randomised dose-finding trial, 60 IVF patients at high risk of ovarian hyperstimulation were given a single kisspeptin-54 injection as the egg-maturation trigger. Eggs matured in 95% of women, the live birth rate per embryo transfer was 45%, and no woman developed moderate, severe or critical OHSS. [23]
- Human studyIn a 2017 brain-imaging study of 29 healthy young men, kisspeptin compared with placebo increased activity in limbic brain areas in response to sexual and couple-bonding images, and reduced negative mood. This was a laboratory study, not a treatment trial. [24]
- Regulator reviewFDA's 2024 review concluded there is insufficient evidence to decide whether kisspeptin-10 works as a treatment for secondary hypogonadism in men. It found no published trial that gave kisspeptin-10 regularly for more than a day; the longest infusion lasted 24 hours. [17]
Kisspeptin side effects and risks
No kisspeptin-10 product is approved in any country, and FDA found too little information to say whether it can be used safely, or at what dose, frequency and duration; it also flagged an unresolved immunogenicity risk for injected kisspeptin-10.
- Regulator reviewAcross the roughly 300 people given kisspeptin-10 in published studies, no serious adverse events were reported. FDA notes these studies were short and small, and often did not report adverse events at all. [17]
- Regulator reviewFDA's adverse-event database holds one report: a 17-year-old with hypogonadotropic hypogonadism who gained weight and had a rise in estrone after six weeks of daily compounded kisspeptin-10 injections. FDA notes the report lacks the detail needed to link the two. [17]
- Human studyRepeated dosing can switch the hormone response off. In women given kisspeptin-54 twice daily, the average peak LH rise after an injection fell from 24.0 IU/L on day 1 to 2.5 IU/L on day 14. [22]
Other things to know
- FDA states kisspeptin-10 'may pose a significant risk for immunogenicity' when injected subcutaneously or intramuscularly, worsened by potential peptide-related impurities and aggregation that the nominator's data did not rule out.
- Nonclinical pharmacology data show tachyphylaxis: uninterrupted dosing can desensitise the KISS1 receptor and render kisspeptin pharmacologically inactive, which is the opposite of the intended hormonal effect.
- FDA found no published studies assessing adverse events at the doses and frequencies actually used to treat reproductive disorders - only short, single- or few-dose studies in healthy volunteers.
- Prohibited at all times for male athletes under the WADA 2026 Prohibited List, section S2.2.1 (testosterone-stimulating peptides), because it can raise LH-driven testosterone.
- FDA notes several already-approved treatments exist for secondary hypogonadism in men, a factor it weighed against adding kisspeptin-10 to the compounding list.
