Kisspeptin Dosage: Common Protocol, Study Doses and Calculator

Kisspeptin dosage: the 50-200 mcg common protocol, the IV bolus and twice-daily study doses that are actually published, and a calculator.

The short version

Kisspeptin has no approved dose because no kisspeptin product is FDA-approved. Dosing-guide sources report a common protocol of 50-200 mcg by subcutaneous injection once or twice daily, in 1-6 week cycles. None gives a break period. This is labelled common practice, not trial-established. Published human studies used IV bolus doses as small as 0.01 microgram/kg. One used a twice-daily subcutaneous dose of 6.4 nmol/kg for two weeks, which had stopped working by day 14.

  • Dosing-guide sources report 50-200 mcg subcutaneous, once or twice daily, in 1-6 week cycles, with no break period. This is common practice, not an established dose.
  • Published human dosing comes from short, monitored infusion and injection studies, not a repeated take-home protocol.
  • The one study that tried twice-daily dosing for two weeks found the hormone response had mostly disappeared by the last dose.
  • Higher isn't better: a 3 microgram/kg IV bolus produced a smaller LH rise than 1 microgram/kg in the same study.

Kisspeptin has no approved dose, because no kisspeptin product has completed FDA review. This page reports what two published human studies gave their own volunteers under monitoring. It is not a recommendation for what anyone should take. For the fuller research record and legal status, see the main kisspeptin guide.

US legal status - Research use only

No kisspeptin product is FDA-approved. FDA's October 2024 briefing document proposed NOT adding kisspeptin-10 to the 503A Bulks List, so it is not presently eligible for lawful 503A compounding for the use FDA evaluated.

Kisspeptin dosage: the common protocol

The protocol most often used is 50-200 mcg by subcutaneous injection once or twice daily, run in 1-6 week cycles. No source gives a break period between cycles. This is common practice, not a trial-established or FDA-approved dose.

Common protocol

What is commonly used, not a dose established in trials. No regulator has set a dose for Kisspeptin.

The 50-200 mcg flat-dose, cyclical protocol above is a community research convention for kisspeptin-10 - it has not been tested in a controlled trial. Controlled human studies instead use kisspeptin-54 dosed by body weight for reproductive-medicine purposes: a single 6.4-12.8 nmol/kg subcutaneous injection to trigger oocyte maturation before IVF egg retrieval, or twice-daily 6.4 nmol/kg subcutaneous injections for 7 days (menstrual cycle days 7-14) in healthy women, which did not abolish menstrual cyclicity - though the same trial notes chronic kisspeptin exposure has caused desensitization in other populations (male monkeys, women with hypothalamic amenorrhea). Neither trial tested the multi-week, flat-mcg regimen above, so its cycle length and dose remain a community convention; no source gives a specific break period, so none is stated here.

Plan a cycle in the protocol builder or work out injection volume in the dosage calculator.

The common Kisspeptin dose is 50-200 mcg, once or twice daily, 50-200 mcg per injection. From a 10 mg vial mixed with 2 mL of bacteriostatic water, 150 mcg is 3 units (0.03 mL) on a U-100 insulin syringe.

Kisspeptin doses used in human studies

Two study designs make up the core of the published human dosing record. They used two different populations and two different kisspeptin forms.

Male bolus study. Kisspeptin-10 raised serum LH from about 4.1 to 12.4 IU/liter at the 1 microgram/kg dose within 30 minutes. The higher 3 microgram/kg dose produced a smaller rise than 1 microgram/kg in the same men.

Male infusion study. The lower-rate infusion (1.5 micrograms/kg/hour) raised LH pulse frequency. The higher-rate infusion (4 micrograms/kg/hour) raised LH further but obscured the normal pulsing pattern. Testosterone rose alongside it.

Female trial. The twice-daily subcutaneous schedule produced a strong LH and FSH rise after the very first injection (mean maximal increase: LH 24.0 IU/liter, FSH 9.1 IU/liter). By the 14th injection day, the same dose produced a far smaller rise (LH 2.5 IU/liter, FSH 0.5 IU/liter). The authors attribute this statistically significant drop to tachyphylaxis, a rapid loss of response to repeated dosing.

Why more or more often is not the answer here

Two findings point the same way. In the male study, 3 micrograms/kg produced a smaller LH rise than 1 microgram/kg, so a higher single dose did not mean a bigger response. In the female study, the identical twice-daily dose that worked strongly on day one had mostly stopped working by day 14. Raising the dose or the frequency, based on these two studies, risks blunting the hormone response rather than amplifying it. That is the opposite of what a repeated-dosing plan would assume.

What is not established

  • No approved dose exists, for any population, indication, route or duration, because no kisspeptin product has completed FDA review.
  • No dosing schedule longer than two weeks has been published. FDA’s own review found the longest continuous human infusion was 24 hours. The longest repeated-dose study was the two-week twice-daily trial above, which ended with a weakened response.
  • No human half-life supports a spacing recommendation beyond what these studies used. Kisspeptin-10 has a reported IV half-life of about 3.8-4.1 minutes in the studies FDA reviewed. FDA notes the concentration-time profile differs for a subcutaneous injection, and it does not describe a vial dosed at home.
  • The common protocol above is not the trial protocol. It reflects what dosing-guide and community sources describe as typical practice, not a dose any study or regulator has established as safe or effective.

Reconstitution and injection volume: use the calculator, not a guess

Turning a target amount into an injection volume depends on how a specific vial was reconstituted: how much diluent, and what resulting concentration. This page cannot know that for your vial. The dosage calculator walks through vial strength, diluent volume and target amount. It performs the math. It does not tell you what dose to use.

Who should not use it

No FDA-reviewed contraindication list exists, because kisspeptin has never gone through that review for any consumer use. FDA’s 2024 evaluation found no published study assessing adverse events at the doses and frequencies that treating a reproductive disorder would require. The studies were short, single- or few-dose, in healthy volunteers or small, closely monitored patient groups. There is no published data on pregnancy, breastfeeding, adolescents, or use alongside other medications.

Kisspeptin side effects and risks

FDA states kisspeptin-10 “may pose a significant risk for immunogenicity” when injected subcutaneously or intramuscularly. Unresolved questions about peptide-related impurities and aggregation make that concern worse.

The tachyphylaxis finding above means a repeated schedule can lose effect rather than build one. That is a practical risk for anyone assuming more dosing means more result. Material sold as a research chemical also carries no regulator-verified purity, identity or concentration.

Common questions

What is the usual kisspeptin dose?

Dosing-guide and community sources describe a common protocol of 50-200 mcg by subcutaneous injection, once or twice daily, in 1-6 week cycles. No source gives a break period between cycles. That is common practice reported outside any trial. No regulator or published study has established it as safe or effective. The published human trial data below suggests repeated dosing without a break can stop working.

What dose of kisspeptin have human studies used?

Two study types make up most of the published human dosing record. In healthy men, intravenous bolus doses of 0.01-3.0 micrograms/kg were tested, with the largest LH rise at 1 microgram/kg. The same men also had continuous IV infusions at 1.5 or 4 micrograms/kg per hour for up to 22.5 hours. In women with hypothalamic amenorrhea, a separate trial used twice-daily subcutaneous injections of kisspeptin-54 at 6.4 nmol/kg for two weeks.

Is there an FDA-approved kisspeptin dose?

No. FDA has approved no kisspeptin or kisspeptin-analogue product for any indication. Its own 2024 review proposed against adding kisspeptin-10 to the list of substances pharmacies may legally compound. The figures on this page are what specific small studies used in their own participants, not a recommendation.

Does taking kisspeptin more often work better?

No published study supports that. A trial that gave women twice-daily subcutaneous kisspeptin-54 for two weeks found the LH and FSH response was strong on day one and significantly reduced by day 14. This rapid loss of effect is called tachyphylaxis. Separately, a higher single IV dose (3 micrograms/kg) produced a smaller LH rise than a lower one (1 microgram/kg) in the same men.

How do I work out reconstitution and injection volume?

That is arithmetic about a specific vial and diluent, not a dosing decision, and this page cannot know your vial. Use the dosage calculator to work through concentration and draw volume for whatever amount you are looking at. It does the math. It does not tell you what dose to use.

Who should not use kisspeptin?

No FDA-reviewed contraindication list exists, because kisspeptin has never gone through that review for any consumer use. FDA's 2024 evaluation found no published studies assessing adverse events at the doses and frequencies needed to treat a reproductive disorder. The studies were short and mostly single-dose, in healthy volunteers or small patient groups under close monitoring.

What are the main risks?

FDA's review flagged a potential immunogenicity risk for injectable kisspeptin-10, worsened by unresolved impurity and characterisation questions. The tachyphylaxis findings also mean a repeated dosing schedule can stop working rather than build an effect. Material sold as a research chemical carries no regulator-verified purity, identity or concentration.

When should I stop and get medical care?

Get emergency care (call 911 in the US) for a severe allergic reaction, chest pain, fainting, or any symptom that feels dangerous. If you have taken an unknown or excessive amount of anything sold as kisspeptin, call Poison Control at 1-800-222-1222. None of this is a substitute for a clinician who knows your history.
