Pinealon 10 mg vial dosage

A 10 mg Pinealon vial mixed with 1 mL of bacteriostatic water gives 10 mg/mL, so 1.5 mg is 15 units (0.15 mL) on a U-100 insulin syringe. At 1.5 mg a dose, one vial holds 6 doses.

What Pinealon is

Class:
Synthetic tripeptide (Glu-Asp-Arg)
Also called:
EDR peptide, Glu-Asp-Arg
Used for:
Sold for cognition, focus and healthy aging.
Human evidence:
Limited human data. Twenty-two PubMed records in total, one tagged as a clinical trial and none as a randomised controlled trial. Several records returned under a human-tagged search are false positives about unrelated subjects. Most of what exists is mechanistic laboratory work; the four human studies in my registry are small Russian reports (150 lorry drivers and locomotive crews in 2012, about 30 adults in 2015, 110 people in 2016) whose abstracts describe no placebo group; the only stated dose is 100 mcg by mouth twice daily for two weeks, and the 2015 study reported pro-oxidant activity and a fall in blood stem cells.
US status:
Research use only. Not approved for human use and sold as a research chemical.

Supplies for one cycle with 10 mg vials

Counted for the low and high ends of the common protocol, mixing each vial with 1 mL of water. One new syringe per injection.

Arithmetic on the protocol and this vial size. Swabs: two per vial mixed (both stoppers) and two per injection (vial stopper and skin). An opened multi-dose vial has a use-by date (see storage), which on a slow schedule can matter more than the dose count. Plan your own numbers with the protocol builder.

How to mix a 10 mg Pinealon vial

For this vial: 1 mL of bacteriostatic water makes 10 mg/mL, so 1.5 mg is 15 units (0.15 mL) on a U-100 insulin syringe. Pick a different volume from the units table if you want bigger or smaller draws; the number of doses stays the same.

  1. Wipe the rubber stoppers of both the peptide vial and the bacteriostatic water vial with an alcohol swab and let the alcohol dry. [3][4]
  2. With a new sterile needle and syringe, draw up the amount of bacteriostatic water you have chosen for the dilution. [3][4][5]
  3. Inject the water into the peptide vial by aiming the stream against the inside glass wall, which helps prevent foaming. [6]
  4. Swirl or roll the vial gently until the powder has fully dissolved; do not shake it. Do not inject the solution if it is cloudy or has particles in it. [3][4][6]
  5. Label the vial with the date and time it was mixed, the amount of water added and how long it can be used after mixing. [3]
  6. Store the mixed vial in the refrigerator at 2-8°C (36-46°F) and do not freeze it. [6]

Step by step with pictures: how to reconstitute peptides. About the water itself: bacteriostatic water.

Storing Pinealon vials

  • Freeze-dried (lyophilized) peptide keeps far longer than peptide in solution; peptide suppliers advise storing the powder at -20°C away from bright light. [7]
  • Protect peptide vials from heat and light. [7][8]
  • Avoid repeated freeze-thaw cycles; a prescription peptide label also says not to freeze a vial once it has been mixed. [6][7]
  • Once a multi-dose vial, such as bacteriostatic water, has been opened (needle-punctured), date it and discard it within 28 days unless its manufacturer states another date. [9][10]

More detail: peptide storage and shelf life.

Injecting safely

  • Clean the injection site with an alcohol swab and let the skin dry completely before injecting. [11][12]
  • Pinch up a fold of skin and fat (not muscle) and insert the needle at a 45- to 90-degree angle: 90 degrees where there is enough fatty tissue, 45 degrees where there is little. [11][12][13]
  • Pulling back on the plunger (aspiration) is not necessary for a subcutaneous injection, because the chance of the needle being in a blood vessel is small. [11]
  • Inject slowly, at roughly 10 seconds per mL. [11]
  • Common sites are the belly, the thigh and the upper arm; rotate sites, keeping each injection at least 1 inch (2.5 cm) from the last. [6][11][12]

Hygiene and warning signs

  • Use a new sterile needle and syringe for every injection and every time you enter a vial; never reuse them. [5][9][12]
  • Never share needles, syringes or pens with anyone, even if the needle is changed; unsafe injection practices spread hepatitis B, hepatitis C and HIV. [5][8][9]
  • Put used needles and syringes in a sharps disposal container straight away (FDA-cleared if possible), never loose in the trash, recycling or toilet, and dispose of the container following your community's guidelines. [14][15]
  • Contact a health care provider if the injection site shows signs of infection, such as redness, swelling, red streaks or a fever. [12]

Where to inject: subcutaneous injection sites.

How Pinealon works

  • Pinealon is a synthetic three-amino-acid peptide, Glu-Asp-Arg (EDR), developed by Vladimir Khavinson's peptide-bioregulator group at the Saint Petersburg Institute of Bioregulation and Gerontology. [16][17]
  • In cultured cerebellar granule cells, neutrophils and PC12 cells under oxidative stress, pinealon limited the build-up of reactive oxygen species in a dose-dependent way and reduced necrotic cell death, alongside delayed ERK1/2 activation and changes in the cell cycle. [16]
  • Fluorescently labelled pinealon entered the nucleus of cultured HeLa cells and bound preferentially to certain DNA sequences in test-tube experiments. Its developers propose that it binds histones or RNA and so changes the production of proteins such as caspase-3, p53 and antioxidant enzymes. [17][18]

What the Pinealon research found

Human evidence: limited human data. Each result says what was studied.

  • Animal studyPregnant rats fed extra methionine to raise homocysteine had offspring with better spatial learning, and less reactive-oxygen build-up and fewer dead cells in neurons taken from their cerebellum, when the mothers were given pinealon. [19]
  • Lab studyA 2020 review by the developers reports that EDR reduced the loss of dendritic spines in neuronal cultures from mouse models of Alzheimer's and Huntington's disease. [17]
  • Human studyIn a 2015 Russian study of 32 adults aged 41-83 with multiple chronic conditions and organic brain syndrome, pinealon and a related peptide, vesugen, were reported to slow measures of biological age, with vesugen showing the larger effect. The abstract does not describe a control group, and its own breakdown of 18 men and 12 women adds up to 30. [20]
  • Human studyA 2012 Russian study of professional lorry drivers reported improved psychological and emotional scores after bioregulator peptides, with the best result from pinealon combined with vesugen. The abstract does not say how many drivers received peptides or whether a placebo was used. [21]
  • Human studyA 2012 Russian study gave workers on locomotive crews one 100 mcg Pinealon capsule by mouth twice a day for two weeks and reported improved biological-age and adaptation measures; the abstract gives no participant count and no comparison group. [22]
  • Human studyA 2016 Russian comparison of several anti-ageing methods in 110 people found that Vesugen combined with Pinealon had the most marked effect on biological-age indicators; the abstract does not describe randomisation or doses. [23]

Pinealon side effects and risks

I found no controlled trial with systematic adverse-event data for pinealon; the human reports I cite are a few small Russian-language studies whose abstracts give little safety detail.

  • Human studyIn the 2015 study of 32 adults, the authors recorded pro-oxidant activity on chemiluminescence testing and a fall in blood CD34+ stem cells, which they described as significant inhibition of blood-cell formation. The abstract does not separate the effects of pinealon and vesugen. [20]
  • Human studyThe same authors reported that pinealon and vesugen did not change chromatin condensation, which they interpreted as safety at the level of the cell nucleus while saying this needs further study. [20]
  • Human studyThe 2016 comparison rated massage and the peptide preparations (Vesugen and Pinealon) as the safest of the methods tested on biochemical, immune and clinical measures; the abstract gives no adverse-event counts. [23]

Other things to know

  • There is no controlled clinical evidence - the human studies I could check are small, reported in Russian, and describe no placebo group.
  • A 2015 study of adults given pinealon and vesugen reported pro-oxidant activity and a fall in blood CD34+ stem cells, which its authors called significant inhibition of blood-cell formation.
  • No dose has research behind it, so any protocol was produced by someone other than a researcher.

Questions

How much water do I add to a 10 mg Pinealon vial?
There is no single correct amount: water changes how many units a dose is, not how many doses the vial holds. With 1 mL the vial is 10 mg/mL and 1.5 mg is 15 units (0.15 mL) on a U-100 insulin syringe. The units table gives 1, 2, 5 mL.
How many doses are in a 10 mg Pinealon vial?
10 doses at 1 mg, 6 doses at 1.5 mg, 5 doses at 2 mg.
How long does a 10 mg Pinealon vial last?
At 1.5 mg a dose and once daily, about 0.9-1.2 weeks. How long a mixed vial keeps is a separate limit; see storage below.
How many 10 mg vials do I need for a cycle?
1 vial at 1 mg 5 times a week for 1.4 weeks; 5 vials at 2 mg 7 times a week for 3 weeks.
What does 1 unit equal in a 10 mg Pinealon vial?
100 mcg with 1 mL, 50 mcg with 2 mL, 20 mcg with 5 mL of bacteriostatic water, on a U-100 syringe.
