MK-677 Cycle Length: How Long People Run It vs Trials
MK-677 cycle length: the 8-12 week common protocol with a 4-week break, contrasted with trials that ran 14 days to 2 years continuously.
The short version
The common protocol people describe for MK-677 is 12.5-25 mg by mouth once daily for 8-12 weeks, then a 4-week break. That is common practice, not a cycle any trial tested. The published human trials ran differently: 14 and 28 days in one dose-ranging study, 8 weeks in two others, and 2 years continuously with no break in the longest. That trial reported transient lower-leg edema and an appetite increase that subsided within months for about half of subjects. Fasting glucose rose and insulin sensitivity fell, significantly at 12 months but not in subjects followed to 24 months. It is the closest published evidence on continuing past a single cycle.
- The common practice cycle is 8-12 weeks on, 4 weeks off. It is not trial-established.
- No published trial cycled MK-677. The longest ran 2 years continuously with no break.
- In that 2-year trial, edema and appetite were transient for most subjects. Glucose and insulin-sensitivity changes were significant at 12 months but not at 24.
- Whether a 4-week break changes any of that has not been tested in a published trial.
MK-677 has no FDA-approved dose or cycle, because it has no approved product. This page contrasts the cycle length people commonly describe with how long the published human trials ran, and what those trials reported changing over time. For the dose itself and full trial detail, see MK-677 dosage.

MK-677 cycle length: the common protocol
Community and clinic dosing guides most often describe 12.5-25 mg by mouth once daily for an 8-12 week cycle, followed by a 4-week break before starting again. This is common practice, not a cycle length any trial has tested. See MK-677 dosage for the full sourcing behind those numbers and the protocol builder to lay it out against other compounds.
How long people run it vs how long the trials ran
No published human trial of MK-677 cycled it the way common practice describes. The longest trial ran for 2 years straight, with primary endpoints assessed at 1 year. That is more than eight times longer than even the top end of the common 12-week cycle, with no break built into the design at all.
Water retention, appetite and glucose over the course of use
Only the 2-year trial is long enough to show how effects change over time, so this section leans on it. The shorter trials are used where they overlap.
- Water retention (edema). The 2-year trial recorded transient lower-leg edema in 44% of subjects on MK-677 vs 27% on placebo, one of the most frequent reported effects Nass 2008 - Nass R, Pezzoli SS, Oliveri MC et al. Effects of an oral ghrelin mimetic on body composition and clinical outcomes in healthy older adults: a randomized trial. Ann Intern Med. 2008;149(9):601-11., in the references below. It describes the edema as transient but does not report exactly when it resolved, or whether a break changes that.
- Appetite. Increased appetite was the most frequently reported effect, in 67% of subjects on MK-677 vs 36% on placebo Nass 2008 - Nass R, Pezzoli SS, Oliveri MC et al. Effects of an oral ghrelin mimetic on body composition and clinical outcomes in healthy older adults: a randomized trial. Ann Intern Med. 2008;149(9):601-11., in the references below. The trial describes it as subsiding within a few months in about half of those subjects, and more gradually in others. It is not described as persisting unchanged for the full 2 years.
- Fasting glucose. In the short dose-ranging trial, fasting glucose rose from 5.4 to 6.8 mmol/L by 4 weeks on the 25 mg dose Chapman 1996 - Chapman IM, Bach MA, Van Cauter E et al. Stimulation of the growth hormone (GH)-insulin-like growth factor I axis by daily oral administration of a GH secretogogue (MK-677) in healthy elderly subjects. J Clin Endocrinol Metab. 1996;81(12):4249-57., in the references below. The 2-year trial reported a smaller rise, about 0.3 mmol/L, and reduced insulin sensitivity, both significant at the 12-month primary endpoint Nass 2008 - Nass R, Pezzoli SS, Oliveri MC et al. Effects of an oral ghrelin mimetic on body composition and clinical outcomes in healthy older adults: a randomized trial. Ann Intern Med. 2008;149(9):601-11., in the references below. In the subjects who continued through the full 24 months, that trial’s own exploratory analysis found fasting glucose was no longer significantly above baseline. Its data do not show a rise that simply continues unbroken with longer use.
- Oral glucose tolerance. In the 8-week obese-men trial, oral glucose tolerance was impaired at both the 2-week and 8-week measurements Svensson 1998 - Svensson J, Lönn L, Jansson JO et al. Two-month treatment of obese subjects with the oral growth hormone (GH) secretagogue MK-677 increases GH secretion, fat-free mass, and energy expenditure. J Clin Endocrinol Metab. 1998;83(2):362-9., in the references below. It was present early and still present at the end of that shorter trial.
Should you cycle MK-677 or take it continuously?
Common-practice sources recommend an 8-12 week cycle with a 4-week break. They commonly reason that a break allows labs and physician review between courses. No published trial compared cycled to continuous use, so that reasoning is common practice, not a tested finding. The dosing page makes the same distinction about the dose itself. The only long-duration data available is the 2-year continuous trial above. It answers a different design question (continuous use, no break) and is not evidence for or against cycling.
What is not established
- No trial tested cycled use. Every duration figure above comes from a continuous or fixed-period design, not a cycle-and-break structure.
- No trial tracked the same people from a short cycle through a break and a second cycle. The 2-year trial is the closest long-duration data available, but it used one continuous 2-year exposure, not repeated cycles.
- Whether effects reset during a break has not been studied. Nothing here shows what happens to appetite, edema or glucose during the 4-week break common practice describes.

Common questions
How long should an MK-677 cycle be?
Community and clinic dosing guides most often describe an 8-12 week cycle at 12.5-25 mg by mouth once daily, followed by a 4-week break before starting again. That is common practice reported by real dosing sources, not a cycle length any published trial tested.
Did the human trials of MK-677 use cycles?
No. None of the four published human trials cycled MK-677. A dose-ranging trial ran two separate periods of 14 and 28 days. Two trials in obese men ran a continuous 8 weeks. The longest, in healthy older adults, ran 2 years continuously with no defined break.
Does MK-677 water retention go away over time?
The only trial long enough to speak to this, a 2-year study in older adults, described the lower-leg edema (swelling) it recorded as transient. That means it did not persist for the full 2 years. No trial tracked exactly when swelling resolved or whether a break changes that pattern.
Does blood sugar keep rising the longer you take MK-677?
Two trials measured it over different lengths. In a dose-ranging trial, fasting glucose rose within 4 weeks. The 2-year trial reported a smaller fasting glucose rise and reduced insulin sensitivity, both significant at the 12-month mark. In subjects who continued through the full 24 months, that trial's own analysis found fasting glucose was no longer significantly above baseline. So its data do not show the rise simply continuing unbroken through 2 years. No trial tested whether a break changes any of this.
Is there evidence that taking a break from MK-677 helps?
No published trial tested cycling MK-677 against continuous use, so there is no trial evidence either way about whether a break changes appetite, water retention, glucose or any other effect. The 4-week break in common practice comes from dosing guides, not from a study that compared cycled and continuous use.
What is the longest a human trial has given MK-677?
2 years, continuously, with primary endpoints assessed at 1 year, in 65 healthy adults aged 60-81. No published trial has studied use beyond that window, cycled or continuous.
