Humanin Dosage: Common Protocol, Mouse Doses and Calculator
Humanin dosage: the 0.5-5 mg common protocol, the mouse doses from two published HNG-analog studies, and a calculator for injection volume.
The short version
No completed human trial has given anyone humanin, so there is no trial-established dose. The protocol most often used is 0.5-5 mg of native humanin by subcutaneous injection, once daily or a few times a week, for 4-12 weeks with a break between cycles. That is common practice, not a tested or approved dose. The only trial-grade numbers come from two published mouse studies of a modified analog called HNG, not native humanin.
- No completed human trial has ever administered humanin, at any dose.
- The protocol most often used is 0.5-5 mg SC of native humanin, once daily to a few times weekly. It is common practice, not trial data.
- The published mouse studies used an analog called HNG, dosed far differently from native humanin.
- There is no accepted method for converting a mouse dose into a human dose.
No completed human trial has ever given a person an injection of humanin, so nothing below is a trial-established dose. This page gives the protocol most often used in practice, as common practice sourced to the vendors and community guides that publish it. It also gives the only trial-grade numbers that exist, from two published mouse studies of a modified analog, not the native peptide. For the human evidence and legal status, see the main humanin guide.
US legal status - Research use only
Humanin has no FDA-approved product and no dose established by any regulator or trial. It is sold online as a research chemical, not manufactured or labelled for human use.
Humanin dosage: the common protocol
The protocol most often used is 0.5-5 mg of native humanin by subcutaneous injection, once daily or a few times a week, for 4-12 weeks with a break between cycles. Vendors and community guides report this. No trial has tested it.

These figures are for the native humanin peptide sold online, the form these sources describe protocols using. One source separately describes a synthetic analog, HNG (S14G-humanin), dosed roughly 1,000 times lower at 5-10 mcg because of its much higher potency - a different substance from native humanin, not reflected in the range above.
Plan a cycle in the protocol builder or work out injection volume in the dosage calculator.
The common Humanin dose is 0.5-5 mg, commonly once daily; some protocols use 2 to 3 times a week instead. From a 5 mg vial mixed with 1 mL of bacteriostatic water, 3 mg is 60 units (0.6 mL) on a U-100 insulin syringe.

Units are on a standard U-100 insulin syringe (100 units = 1 mL), rounded to the nearest half unit - half-unit marks are on 0.3 mL syringes. Water volumes are examples: more water means more units per dose, not more doses per vial. For any other vial or dose, use the calculator below.

What the human record contains: nothing administered
As the main guide explains, every human study located for this topic measures the humanin a person’s own body already produces, correlating blood levels with age or disease. Not one gives anyone an injection of humanin. There is no completed human trial to draw a dose from.
Humanin doses used in animal studies
Two published mouse studies give real dosing numbers. Both used HNG (S14G-humanin), a laboratory-modified analog engineered to resist the rapid enzymatic breakdown that limits the native 24-amino-acid peptide. A dose of this engineered, more stable analog is not the same substance as the peptide sold online as “humanin.”

The obesity study reported that HNG significantly altered amino-acid and lipid metabolites in plasma over a short, 3-day course. The aging study reported that 14 months of HNG reduced collagen buildup and fibrosis-related signalling in the hearts of aged mice, compared with untreated aged controls. Both are experimental results in mice, using an analog, at the doses and schedules stated in their own methods sections.
What is not established
- No human dose exists, at any route, frequency or duration, because no completed human trial has administered humanin, or its analog HNG, to anyone.
- No human half-life is established. The main guide explains why this site does not publish one. So there is no evidence-based basis for any dosing interval.
- The common protocol above is not trial-established. It reflects what vendors and community guides report using, not a dose any completed trial has tested or any regulator has reviewed.
Reconstitution and injection volume: use the calculator, not a guess
The injection volume for a target amount depends on a specific vial’s concentration and diluent, which this page cannot know. Doing that math changes nothing about the fact that no dose is established. The dosage calculator handles concentration and draw-volume arithmetic; it does not supply or endorse a dose.
Who should not use it
No FDA-reviewed contraindication list exists, because neither humanin nor HNG has ever gone through that review in humans. There is no published human safety data on pregnancy, breastfeeding, children, drug interactions, or use alongside any diagnosed condition. A peptide with no completed human trial and no FDA-reviewed label is not something to self-direct dosing around.
Humanin side effects and risks
The biggest risk is the size of the gap in the data. No human trial has ever administered humanin or HNG, so no human safety profile, human half-life or human dose exists. Material sold online as a research chemical also carries no regulator-verified purity, identity or concentration. That is a safety consideration independent of any effect the peptide might have.

Common questions
What is the usual humanin dose?
No completed human trial has tested a dose, so nothing here is trial-established. Vendors and community sources most often describe 0.5-5 mg of native humanin by subcutaneous injection. The schedule is once daily or a few times a week, for 4-12 weeks with a break between cycles. That is common practice, not a tested or approved dose.
What dose have animal studies used?
Two published mouse studies give concrete numbers. Both used a modified, more stable analog of humanin called HNG (S14G-humanin), not the native 24-amino-acid peptide. One gave diet-induced obese mice 2.5 mg per kilogram by intraperitoneal injection, twice daily, for 3 days. The other gave aging mice 4 mg per kilogram by intraperitoneal injection, twice weekly, for 14 months. These are mouse doses in specific experiments, not a human protocol.
Is humanin the same as the HNG used in these studies?
No. HNG is a laboratory-modified analog of humanin, changed to resist the rapid breakdown that affects the native peptide. Most animal dosing studies use HNG for that reason. A dose of HNG is not automatically a dose of humanin.
Can a mouse dose be converted into a human dose?
Not reliably, and this page will not do it. Body-surface-area scaling formulas exist for regulatory use in early drug development. Applying one here would produce a number with false authority, for a peptide analog that has never been dosed in a human trial.
How do I work out reconstitution and injection volume?
That is arithmetic about a specific vial and diluent, not a dosing decision. It does not change the fact that no dose is established. The dosage calculator handles concentration and draw-volume math for any amount; it does not supply or endorse a dose.
Who should not use humanin, and what are the risks?
With no completed human trial of humanin or HNG, there is no regulator-reviewed contraindication list and no human safety data on interactions, pregnancy, or long-term use. Material sold online as a research chemical also has no verified purity, identity or concentration, whatever the peptide itself does.
When should I stop and get medical care?
In the US, call 911 for a severe allergic reaction, chest pain, fainting, or any symptom that feels dangerous. Call Poison Control at 1-800-222-1222 (verify this number at poisonhelp.hrsa.gov or poison.org) if you have taken an unknown or excessive amount of anything sold as humanin. This is not a substitute for a clinician who knows your history.
