PNC-27: What the Cell-Culture Research Actually Shows
PNC-27 has never been tested in a human. Cultured-cell studies of its cancer-cell-targeting mechanism, and why nobody should self-treat cancer with it.
The short version
PNC-27 is a lab-designed peptide studied for its proposed ability to kill cancer cells selectively. It binds a protein (HDM-2) that researchers find on the surface of cancer cells but not normal ones. All of that evidence comes from cells in a lab dish, not from animals with cancer or from people. It is sold online directly to people with cancer, which is a serious safety concern.
- PNC-27 has never been tested in a human being, in any trial, for any purpose.
- Both published studies used cancer cells grown in a lab, not animals with tumours or people.
- It is marketed to people with cancer, where delaying proven treatment can cost time that matters.
- No dose, safety margin or human protocol has ever been established for it.
PNC-27 at a glance
- What it is
- Synthetic 32-residue chimeric peptide: a p53-derived HDM-2 (MDM2)-binding domain fused to an Antennapedia cell-penetrating sequence
- Main use
- Laboratory and animal-model research into a proposed cancer-cell-selective killing mechanism. No human trial of any kind has been conducted. Sold online, directly to consumers, as an unregulated research chemical marketed against cancer - a serious, life-threatening disease that this compound has never been shown, in any human, to treat.

Dosing: why no dose exists, and what to do instead.
PNC-27 has never entered a human clinical trial. All the evidence behind it comes from cancer cells growing in a lab dish. The gap between “kills cells in a dish” and “treats cancer in a person” is the most important thing to understand about it.
Studied in cells or animals. No controlled trials in humans. Two published studies from the same US academic lab describe PNC-27's mechanism, entirely in cultured human cancer cells. Neither is an animal tumour study or a human trial. I could find no registered human trial of PNC-27.
What is PNC-27?
PNC-27 is a synthetic hybrid peptide of 32 amino acids. One part is the section of the p53 tumour-suppressor protein that binds HDM-2 (also written MDM2). The other is a cell-penetrating sequence from the Antennapedia protein, which helps the peptide cross a cell membrane.

How PNC-27 works, according to the research
HDM-2 normally works inside the cell, where it regulates p53. Researchers report that it also sits on the outer membrane of a range of cultured cancer cells, but not on several normal cell lines they tested (Sarafraz-Yazdi 2010 - Sarafraz-Yazdi E, Bowne WB, Adler V et al. Anticancer peptide PNC-27 adopts an HDM-2-binding conformation and kills cancer cells by binding to HDM-2 in their membranes. Proc Natl Acad Sci U S A. 2010;107(5):1918-23., in the references below).
In these experiments, PNC-27 bound to that surface HDM-2 and punched pores in the cancer cell’s membrane, killing the cell (necrosis). When the researchers made normal cells carry HDM-2 on their surface, those cells became vulnerable to PNC-27 too. That suggests, in a dish, that surface HDM-2 is what the peptide reacts to.
A follow-up study from the same group (Krzesaj 2024 - Krzesaj P, Adler V, Feinman RD et al. Anti-Cancer Peptide PNC-27 Kills Cancer Cells by Unique Interactions with Plasma Membrane-Bound hdm-2 and with Mitochondrial Membranes Causing Mitochondrial Disruption. Ann Clin Lab Sci. 2024;54(2):137-148., in the references below) reports that PNC-27 also gets inside cancer cells and damages the membranes of their mitochondria. The team used an antibody against HDM-2’s binding site, which blocked the effect, plus dyes and electron microscopy to track where the peptide went.
PNC-27 dosage
No dose exists for any purpose, because no human has been given PNC-27 in a trial. Research-market pages that quote a dose are not reporting anything measured. Full sourcing: PNC-27 dosage.
PNC-27 side effects
There is no human safety data at all: no Phase 1 trial and no published case report. The research describes the intended killing of cancer cells in culture, not how people tolerate it.
Is PNC-27 legal?
US legal status - Unapproved drug
Not FDA-approved for any use. I could find no registered human clinical trial. Sold online as an unregulated research chemical, not a medicine.
- No FDA approval, and no disclosed path toward one. There is no registered human trial of PNC-27.
- Not a dietary supplement. It is sold entirely outside the FDA-regulated supply chain, so no regulator checks that a vial matches its label for identity, purity, sterility or amount.
- Even vendors warn against it as a cancer treatment. They routinely tell buyers not to replace oncologist-directed treatment with it. Take that warning at face value.

Common questions
What is the usual PNC-27 dose?
There is no established dose for any purpose, because no human trial of PNC-27 exists. The dosing page shows what real sources say, and where they contradict each other.
What are the side effects of PNC-27?
None are documented in humans, because no human has been given it in a study. The research describes its intended effect on cancer cells in a dish, punching pores in the membrane and killing the cell. That says nothing about side effects in people.
